From my experience, no. But this is certainly true on Windows 8.
I'm almost done with the new launcher that - among other things - addresses settings issue on Windows 8. You will be able to apply this new launcher option on Windows 7 too.

Latest news on EP launcher

First I would like to say thank you all for such a huge interest in my EP launcher.
The new launcher should be ready for download at the beginning of February.

Some of the upcoming launcher's changes & features added
New
- Launcher's config options:
Enable Notes Hyperlinks - you can open notes using hyperlinks from within other apps
Add Send To Shortcut - import any file(s) into EP with right-click -> Send To -> .Evernote Portable
Associate Enex Files - import enex (evernote export) files into EP with just a double-click
- 'settings' file now stores EP settings
- Warning message(s) with instructions when you would leave log files on local computer
- Startup splash screen
Added
- Ability to backup and restore 'settings' file
- Support for the current version (4.6.1.7860)
- Support for the versions prior to 4.6.1.7860 - if you are not ready yet to update your Evernote to the latest version

I updated the launcher and first post.
I almost completely rewrote the launcher. I kept all previous features (except 'Running Mode') and added lots of new ones.
Launcher's changes
- (New) Enable Notes Hyperlinks - you can open notes using hyperlinks from within other apps
- (New) Add Send To Shortcut - import any file(s) into EP with right-click -> Send To -> .Evernote Portable
- (New) Associate Enex Files - import enex (Evernote export) files into EP with just a double-click
- (New) Refresh Shell Icons - you can refresh shell icons
- (New) Add [Debug] Menu - you can add '[Debug]' menu to menus
- (New) Compress Files - you can use launcher to compress EP files
- (New) Custom Upx Path - you can specify custom Upx path: absolute path to folder that contains 'upx.exe'
- 'settings.reg' file now stores EP settings
To use your old EP settings with the new launcher, rename 'rs.reg' file to 'settings.reg'.
- Added warning message(s) with instructions when you would leave log files on local computer
- Added ability to backup and restore settings file
- New startup splash screen
- Many other minor improvements

I updated the launcher.
- Added two new options to configure the launcher:
Fonts Folder - specify absolute path(s) to fonts folder(s) that you would like to use. Separate paths with semicolon.
EP Folder Icon - here you can specify icon for the Evernote Portable folder (absolute path to icon that you would like to use).
- Added support for the custom location of the profile files. Use Tools -> Options -> Evernote local files to specify location.
- Improved updating function.
- Many other minor improvements.

Thanks. This launcher is a really great tool for me.

Is there a way to copy the profile/app back to the local installation?

Or, if I want to do this, which files should be copied where? I don't want to just copy the database, but also the login info.

1. Copy everything from the Profile folder into
(XP) %UserProfile% -> Local Settings -> Application Data -> Evernote -> Evernote;
(7,8) %UserProfile% -> AppData -> Local -> Evernote -> Evernote.
2. Import contents of ‘settings’ file into registry (Regedit).
3. Delete ‘EvernotePath’ value in HKCU -> Software -> Evernote -> Evernote registry key (Regedit).

Major update of EP launcher
IMPORTANT: Portable package format has been changed. If you would like to use your existing portable package with the new launcher you must manually convert it.
Step 1: Move 'Evernote.ini', 'settings' and 'settings.bak' files into Profile folder (Evernote Portable -> App -> Profile).
Step 2: Move Profile and upx folders - one level up - into Evernote Portable folder.
Step 3: Rename App folder to En.

- Launcher is now called 'Evernote(P).exe'.
- Launcher config file is now 'Launcher.ini' file in Evernote Portable -> Profile folder.
- EP settings files are in Evernote Portable -> Profile folder.

Whole launcher script has been thoroughly refreshed.
- Added new 'Clip Image (Evernote Portable)' right-click option to Evernote clipper for IE.
- Launcher now supports Chrome and Firefox web clippers. In web clipper's options select:
(Chrome) Open notes in Desktop client;
(Firefox) Send clips to Evernote on your desktop.
To be able to use web browser's clipper with EP, enable 'Enable Notes Hyperlinks' option in 'Launcher.ini' file under Windows Integration section.
Or you can just run EP (while EP is running you are always able to use all options under Windows Integration section).
- New startup splash screen
