HostsOptimizer rearranges the contents of the hosts file, so the DNS Client service can be enabled when there is a large hosts file. It does so with a maximum of 9 host names per single line, all comments removed (includes disabled entries) and duplicate host names deleted.

According to the forum thread (follow the 'website' link), this has now been updated - take note if you're getting a 404 from the download link.
I had real problems with this in Windows 7. I recommend against it.
